AI對抗武漢肺炎！ 英科智能公開百種候選藥物分子

美國時間6日，美國人工智能(AI)藥物研發公司英科智能(Insilico Medicine)宣布，公司以AI技術平台合成並測試了百餘種可能成為武漢肺炎藥物的分子，並公開發表以2019新型冠狀病毒上關鍵蛋白質「3C-樣蛋白脢」(3C–like protease)為標靶的分子結構。

英科智能表示，這些透過AI平台開發出的分子結構，今日已同步發表於其官網，並上傳至由冷泉港實驗室維護的非營利論文資料庫bioRxiv上，與全球相關研發團隊免費共享，期望幫助疫情盡快撲滅。

英科智能創始人兼執行長Alex Zhavoronkov表示，公司自1月28日啟動這項研發計畫後，研究了由北京全球健康藥物研發中心所發布的治療方針，並決定以對冠狀病毒的繁殖極為重要的3C-樣蛋白脢為藥物標靶。

自1月31日起，英科智能以28種不同的機器學習模型，來設計可能與3C-樣蛋白脢結合，並抑制其功能的新分子。

4天後，對於軟體產出的上萬種新分子設計，英科智能進一步使用機器學習中的生成對抗網路(Generative Adversarial Network, GAN)技術，篩選掉例如金屬化合物等化學活性不太可能成為藥物的設計，挑選出符合候選藥物標準的數千種分子。

Zhavoronkov說明，之所以以該蛋白質為標靶，是因為其結構類似某些已被解構的病毒蛋白酶，且上海科技大學的蛋白質結構專家饒子和博士也已對新型冠狀病毒的該蛋白酶研究甚多。

創立於2014年的英科智能，以其AI平台成為許多大型製藥廠藥物開發的後盾，除了1月14日剛宣布與輝瑞(Pfizer)合作外，也已與葛蘭素史克(GlaxoSmithKline)合作藥物開發、與中國藥品和醫療設備開發商藥明康德(WuXi Aptec)建立合作夥伴關係。目前，公司已通過六輪籌集超過5000萬美元募資。

除了英科智能外，密西根州立大學(Michigan State University)的研究團隊，也於上週在bioRxiv發表了一篇以機器學習技術設計新型冠狀病毒候選藥物的論文，並表示其研究也同步分析了部分有潛力對抗新型冠狀病毒的愛滋病藥物。
