阿茲海默症惡化😰?中研院找到 #蛋白質幫兇!

#阿茲海默症 是最常見的 #老年失智症。過去,科學認為阿茲海默症起因於:🧠大腦中的2種蛋白質錯誤堆疊:
1⃣:蛋白質Aβ(Amyloid-β)
2⃣:Tau蛋白(Tau Protein)
最近,中研院基因體研究中心 #陳韻如研究團隊 證實,導致阿茲海默症病情惡化,還有另一個💢蛋白質幫凶:
3⃣:蛋白質TDP-43
👉本研究證實:大腦 #蛋白質TDP-43 錯誤表現後,會與 #Aβ 作用,產生更有毒的中間產物,造成阿茲海默症病情惡化!
👩🔬陳韻如表示,「我們發現TDP-43在阿茲海默症裡扮演多重的角色:一方面會直接與Aβ反應誘發Aβ寡聚體形成,造成 #神經突觸功能障礙,以及記憶受損;另一方面TDP-43也會增加 #腦部發炎反應,讓患者對於空間的記憶能力降低。」
🌎全球約5,000萬名失智症患者中,有將近六、七成都是阿茲海默症病人。發病後會 #記憶力下降、#喪失語言功能、#空間感退化 #常迷路,造成自己及親友的生活負擔。但目前還沒有有效的治療藥物。
📍本研究有助於掌握更多阿茲海默症的發病特徵及機制,未來,可望透過TDP-43,為神經退化疾病尋找🔍診斷及💊治療的新方法!
📍本研究起始於兩位博士後研究學者施耀翔與杜玲嫻博士,並由張婷宇碩士加入團隊協力完成。研究成果已於今(2020)年11月23日發表於《#自然通訊》(Nature Communications)。

Curbing #TDP-43 May Ease the #Alzheimer’s Disease Progression
Neurodegenerative diseases are making more impacts socially and economically as human life expectancy gets higher and higher. The scientific world still doesn’t have many clues regarding these diseases. Dr. Yun-Ru (Ruby) Chen’s research has been circled around the protein TDP-43 and its role in several neuro-degenerative diseases, in a paper published in the journal Nature Communications, they have reported a new insight into how TDP-43 is affecting Alzheimer’s disease.
The team designed and made several antibodies, they will look into a solution in finding a best antibody that can do a preempt act to interact with TDP-43. If less inflammation can be managed, less memory loss of AD patients could be feasible. A synergistic effect to disrupt TDP-43 and Aβ interaction may also ameliorate AD.

防疫研究是當務之急,罕病研究也需要持續地累積。👍
罕病「亨丁頓氏舞蹈症」(Huntington’s Disease)的病因來自於變異的亨丁頓蛋白(mHtt)異常聚集,堆積在腦部破壞神經元,造成患者腦部神經持續性退化。患者發病後手腳不聽使喚,像在跳舞一樣,末期急速退化,平均存活約15至20年,目前仍無法完全治癒。
中央研究院化學研究所黃人則副研究員與前生醫所杜邦憲助研究員組成的研究團隊,設計出「#雙極性胜肽分子」,能夠將一團團聚集的亨丁頓蛋白分散開來。將此分子透過鼻腔給藥至罹病小鼠後,小鼠的運動與認知功能失調明顯得到改善。

黃人則表示,雙極性胜肽分子一端可以辨識變異的亨丁頓蛋白並附著上去,另一端帶正電,可以利用電荷排斥力🧲,打散亨丁頓壞蛋白,減少對神經細胞的毒性及損傷。

With the increase in the aging population, an effective therapeutic regimen is still an unmet medical need for different neurodegenerative disorders. Dr. Joseph Jen-Tse Huang, an associate research fellow at the Institute of Chemistry in Academia Sinica, Dr. Pang-Hsien Tu, a former assistant research fellow at the Institute of Biomedical Science, and their research teams recently report a self-assembled amphiphilic peptide that can inhibit protein aggregation implicated in the pathogenesis of Huntington’s disease (HD). This new finding was published in Advanced Science on January, 2020.

Amyotrophic lateral sclerosis (ALS), is a progressive neurodegenerative disease that affects nerve cells in the brain and the spinal cord. When the motor neurons die, the ability of the brain to initiate and control muscle movement is lost. With voluntary muscle action progressively affected, patients in the later stages of the disease may become totally paralyzed.
肌萎縮性脊髓側索硬化症(ALS, Amyotrophic lateral sclerosis)俗稱為漸凍人症(因從患病開始,患者身體會慢慢無法運動,猶如被漸漸凍結,故名)是一個漸進和致命的神經退行性疾病。起因是中樞神經系統內控制骨骼肌的運動神經元(motor neuron)退化所致。肌肉會逐漸衰弱、萎縮。 最後,大腦完全喪失控制隨意運動的能力。這種疾病並不一定會如老人痴呆症般影響病人的心理運作。相反,患有晚期疾病的病人可一直保持清晰的思維、保留發病前的記憶與同樣的人格和智力。

Thai TV Host Woody Milintachinda went Live on national television to kick off the Global Ice Bucket Challenge and represent Thailand to help raise awareness and funds to support patients suffering from ALS, a neurodegenerative disease that affects nerve cells in the brain and the spinal cord. Thailand has upped the #icebucketchallenge to a massive public event with over a thousand participants at the Central World Square on 22 August 2014 at 8am.
China and Singapore have been challenged and have 7 days to accept and complete the challenge!
This event is now by far the largest ice bucket challenge with the most turn out at one time in the world.
