最近實驗室在做中研院的CRISPR實驗
中研院建議使用 Invtirogen 的 E.coli Stbl3 strain
來放大他們所提供的 lentiviral vector (plasmid)
因為這類的plasmid在DH5alpha中
會有產量低、甚至發生重組(變異)的問題

但是這一株competent cell，一個反應要價$700元左右
波仕特代理的Endura G10 strain，也要$400元左右
(相較於DH5alpha，一管大概$50元...感覺差很多)

最近實驗室可能會大量使用CRISPR
為了節省老闆的錢...
想起以前大學的時候，會自己做勝任細胞
雖然轉型效率不比市售的好

但是想想
頂多把plasmid加多點，或塗多點菌也是可以吧??
↑是這樣嗎? XD

我要transform的東西
不是ligation products
而是一般construct好的plasmids
所以轉型效率對我而言可能不是很重要
(這些勝任細胞的genotype/phenogype才是我要的吧...)


還是之後的問題沒有我想像中的單純? XD
